Background
Activin receptor type 1 (ACVR1, also known as ALK-2) is a bone morphogenetic protein (BMP) type I serine/threonine kinase receptor of the TGF-β superfamily that, upon ligand binding and phosphorylation by cognate type II receptors, propagates signals by phosphorylating SMAD1/5/8 transcription factors to orchestrate embryonic patterning, skeletal development, and postnatal tissue repair, while pathogenic gain-of-function mutations (e.g., R206H) drive the rare congenital disorder fibrodysplasia ossificans progressiva through aberrant chondro-osseous differentiation and also contribute to the oncogenesis of diffuse intrinsic pontine glioma by hyperactivating the BMP–SMAD axis .
